Cl- ion channels have always been underestimated principally due to the difficulties to characterize their molecular identities and by the absence of specific inhibitors. A growing number of evidences suggest that chloride channels dynamically regulated intracellular chloride concentration and play a crucial role in the transmission of cellular signals by controlling oxidative stress level. In this seminar, I will summarize recent advances concerning 2 different chloride channels: CFTR and the VRAC/LRRC8. and their abilities to transport chloride ions but more surprisingly other substrates such as glutathione or ATP. I will also describe our latest findings identifying these 2 chloride channels as major actors in the control of cell destiny and the regulation of oxidative stress level.
